Many of medicine's most powerful drugs are held back by a structural flaw. Rather than masking that flaw, we correct it at its molecular source through chemistry — preserving what makes these drugs effective while removing what makes them harmful.
Our first program targets invasive fungal infections, one of the most critical and least addressed unmet medical needs, with Amfurexin®, a reengineered derivative of the reference antifungal amphotericin B.
Amphotericin B is the broadest-spectrum antifungal and the only one without documented clinical resistance in half a century. Yet a structural flaw limits its use: in aqueous media, the molecule self-aggregates, causing severe toxicity and restricting administration.
Lipid and liposomal formulations have only masked this toxicity through delivery, without correcting its cause — at a high cost.
NS2605 (Amfurexin®) is a new chemical entity derived from amphotericin B. Through a targeted modification, it corrects the molecular root of toxicity — self-aggregation — by dispersing the molecule into a soluble monomeric form.
In vitro proof-of-concept establishes a profile with no equivalent in its class.
